Updating homepage shows 'Page not found'

Whenever I try to update my home page, it results in a "Page not found" page. What may be the problem? Someone please help me, its very urgent!

did you change the permalinks recently?

a new plugin? Does it work with all plugins disabled?

do you have a link to take a look?

Actually, I didn't make any changes recently. Let me try with the plugins disabled.
